Definiens is the pioneer in Tissue Phenomics® solutions for diagnostics development and commercialization and was recently acquired by MedImmune, the biologics research and development arm of AstraZeneca.

As a new customer of microDimensions, Definiens will be combining microDimensions’ co-registration technology with their own technology to enhance the work of their internal research team in immuno-oncology. The goal is to automatically align immunohistochemically stained serial tissue sections with unprecedented speed and accuracy. This, for instance, enables the computation of spatially resolved, highly multiplexed readouts which are required to discover highly predictive diagnostic biomarkers for personalized cancer treatment.

High-Resolution 3D Histology with Voloom 2.4

microDimensions today released a major update of its flagship product Voloom, the fastest software in its class to reconstruct serial sections and facilitate 3D histology. microDimensions Voloom 2.4 can export fully aligned sections in a resolution four times higher than the screen resolution. They can be stored as image series or 3D volumes in the TIFF, IMS (Bitplane Imaris), or MHA (ITK MetaImage) file formats.

Furthermore,

  • it now supports reading Hamamatsu NDPI and Aperio SVS confocal images and Z stacks,
  • handling of electron microscopy images is improved,
  • wrong or missing metadata can be corrected by manually specifying in-plane pixel spacing and background color,
  • the user interface was improved,

Memorial Sloan Kettering Cancer Center Uses Voloom for 3D Reconstruction of Immunofluorescence Serial Sections

In a recent publication, Fujisawa et al. [1] describe how they used Voloom to reconstruct immunofluorescently stained adjacent sections. Apart from visualizing geometric structures, this enables them to study and analyze the presence and interactions of multiple specific proteins and molecules in high resolution (using Imaris from Bitplane). This would have been very challenging in tissue blocks, thick sections, or whole mount samples.

In their study they focused on high automation of the processing, staining, imaging, and alignment of sectioned paraffin embedded samples. Their method enables the visualization, analysis, as well as observation of the clear separation, localization and interaction of multiple antigens, proteins, and molecules in tissue samples with a thickness of more than 100 microns.

microDimensions Zoom now smartly reacts to user interactions. During panning and zooming an overview window shows where the current view is located inside the whole slide image. Bitplane and microDimensions Announce Partnership to Bring Voloom to the Wider Life Sciences Market

Bitplane, an Oxford Instruments company and microDimensions announce a partnership to bring Voloom®, from microDimensions, to the wider life sciences market via the Bitplane sales channels.

Voloom is an intuitive and automated software solution for the alignment and reconstruction of whole slide scanner images (and other series of 2D images) of any size. Voloom enables the examination of tissue slices as a 3D volume thus revolutionizing the way users of whole slide imaging devices can see and interact with the entire 3D sample. Imaris natively reads Voloom output and can determine a number of key parameters (i.e. volume, shape, surface structure and many more quantitative measures).

The combination of Voloom and Imaris enables researchers who typically work with 2D serial sections (e.g. automated whole slide scanner or conventional microtome+microscope users) to complete quantitative analyses on 3D volumes. Initially both bright-field and fluorescence data are supported but in the mid-term more imaging modalities will be added to the workflow.
